 言語は、ある特定の国や地域や文化に属する人々のコミュニケーションや相互作用において使用されるもので、音声や書かれた記号からなる体系である。脳科学が対象とする言語は、言語学の知見、神経心理学の知見、そしてイメージング研究の知見を総合して考える必要がある。言語学の主な分野としては、音韻論、形態論、統語論、意味論、語用論があり、代表的な理論には、生成文法、認知意味論、そして関連性理論がある。神経心理学では、失語症の研究から、環シルビウス溝言語領域が、音韻論、形態論、そして統語論の神経基盤、すなわち言語中枢と考えられている。また、環・環シルビウス溝言語領域は意味論的な情報の充填、右半球言語領域は語用論的な情報の充填に関与することを予想している。イメージング研究では、環シルビウス溝言語領域が言語中枢であること、環・環シルビウス溝言語領域が意味論的な情報の充填に関与していることを支持している。さらに、内側前頭前野および後帯状回は、語用論的な情報の充填に関与していることが示唆されている。なお、語用論的な情報の充填に関しては、右半球言語領域や皮質下領域の関与も示唆されているものの、一貫した見解は得られていない。

言語とは

 言語は、ヒトが音声や文字を用いて思想・感情・意志などを伝達するために用いる記号体系であり(広辞苑, 大辞泉)、ヒトはそれを表現したり、他者のそれを受け入れて理解したりする(大辞泉)。ある特定の国や地域や文化に属する人々のコミュニケーションや相互作用において使用されるもので、音声や書かれた記号からなる体系である。表情や身振り手振りなども含む場合がある。

 脳科学が対象とする言語は、言語学の知見、神経心理学の知見、そしてイメージング研究の知見を総合して考える必要がある(図1)。

 言語学では、音声や書かれた記号を対象として、人々によってそれがどのように使用されるのかを明らかにしようとしている。

 神経心理学では、脳の物理的損傷の患者を対象として、その損傷により引き起こされたと考えられる症状との関係から、脳における言語機能を明らかにしようとしている。

 イメージング研究では、主に健常者を対象として、あるタスクを行ってもらい、その際の脳の賦活部位を、機能的磁気共鳴画像(fMRI: Functional Magnetic Resonance Imaging)などの手法を用いて明らかにしようとしている。

言語学の知見

 言語学では、ヒトが使用する言語を客観的に記述し説明することを目的としており、その分野は多岐にわたる。音韻論は言語の構成要素である音声の機能、形態論は語を構成する仕組み、そして統語論は語が文を構成する仕組みをそれぞれ研究対象としている。また、意味論は語、句、文などの言語表現が表す意味、語用論は言語表現とその使用者(話し手、聞き手、書き手、読み手)や文脈との関係をそれぞれ研究対象としている。

 代表的な理論(言語的モデル)としては、生成文法認知意味論認知言語学の主要な理論)、そして関連性理論を挙げることができる。

  • 生成文法(generative grammar)は[1]、ヒトの発達においてごく短期間に言語獲得が成功することに注目し、生得的に言語の初期状態である普遍文法(universal grammar, UG)を備えていると仮定したモデル化をしている。そのため、新生児が育つ国や文化によってどのような言語でも短期間に獲得できるのは、この生得的な普遍文法によると考えられている。
  • 認知言語学(cognitive linguistics)は[2]、生得的で自動的な認知能力(autonomous cognitive faculty)として言語を捉える生成文法の立場に異議を唱え、動的に概念構成(conceptualization)していくという文法の役割を強調し、ことばの意味は使用を通してあらわれるという仮説を唱えている。研究は、意味論に集中しており、認知意味論(cognitive semantics)は[3]隠喩(metaphor)、換喩(metonymy)、イメージスキーマ(image schema)を用いて言語の実態の究明を目指している。
  • 関連性理論(relevance theory)は[4]関連性という認知効果処理労力のバランスで定まる情報の属性を手掛かりとして、聞き手は「話し手が伝えたいと思っている意味」を推論しているという論を展開している。

 ことばの鎖(speech chain)は[5]、話し手の言語学的な段階(linguistic level)と生理学的な段階(physiological level)から、音響学的な段階(acoustic level)を経て、聞き手の生理学的な段階と言語学的な段階に至るという言語コミュニケーションにおける一連の流れをモデル化している(図2)。話し手は言語産出(speech production)の相、聞き手は言語理解(perception)の相のモデルとなっている。通常の会話では、聞き手は次の話し手となるため、ことばの鎖は循環構造となる。

 意味づけ論(sense making theory)は[6][7]、空気の振動としての音声やインクの染みとしての文字をコトバ(カタカナで表記)と定義し、複数の会話の参加者の情況(mental states)はコトバを介して相互作用するという論を展開している。

神経心理学の知見

 神経心理学では、脳の物理的損傷の患者を対象として、その損傷により引き起こされたと考えられる症状との関係から、脳における言語機能を明らかにしようとしている。言語処理に関与する脳の領域は、大まかには、環シルビウス溝言語領域(言語中枢)、環・環シルビウス溝言語領域、そして右半球言語領域の三領域に分けることができる(図3)[8]

  • 環・環シルビウス溝言語領域(peri-perisylvian speech zone)は、環シルビウス溝言語領域の周りの側頭葉頭頂葉前頭葉を含み、その活動には補足運動野視床も加わり、音声系列への言語的意味の充填に関与していると考えられている。左中下側頭回の変性病巣で語義理解の障害[14]、左側頭葉前方で固有名詞の回収障害[15]が報告されている。また、補足運動野は会話の開始および維持において重要な役割を果たしている可能性[16]、視床は語彙を長期記憶から呼びだして文に組み込む役割を果たしている可能性[17]が示唆されている。
  • 右半球言語領域(right hemisphere language zone)は、状況に応じた言語使用、比喩、談話主題の維持、言語による情動表現など、語用論において重要な役割を果たしている。この領域の傷害により、比喩理解の障害、ユーモア理解の障害、談話の一貫性の消失などが報告されている[18]。また、感情を言語にこめられなくなったり、言葉が含む感情が理解できなくなったりすることも報告されている[19]

 ウェルニッケ=ゲシュビント・モデルでは[20]、言語理解の相と言語産出の相をつなぐことで、ヒトが聞き手ないし読み手として言語理解をしてから、話し手ないし書き手として言語産出をするまでをモデル化している。話し言葉は耳で知覚して、視床の内側膝状体を経由して、大脳皮質上側頭回にある一次聴覚野へ情報が入る。言語中枢であるウェルニッケ野から弓状束を通りブローカ野に至る領域で理解と産出をおこない、そして一次運動野から口を制御して音声を発するという経路をたどる。書き言葉は目で知覚して、視床外側膝状体を経由して、大脳皮質の後頭葉にある一次視覚野へ情報が入る。側頭頭頂接合部にある角回を経由して、言語中枢で理解と産出をおこない、そして一次運動野から手を制御して文字を記すという経路をたどる。

 いずれも、主に環シルビウス溝言語領域における活動のモデル化となっており、言語学における音韻論、形態論、そして統語論の神経基盤、すなわち言語中枢と考えられている。これに、環・環シルビウス溝言語領域による意味論的な情報の充填、右半球言語領域による語用論的な情報の充填がおこなわれていると予想されている[8]

イメージング研究の知見

 イメージング研究では、主に健常者を対象として、あるタスクを行ってもらい、その際の脳の賦活部位を、機能的磁気共鳴画像や陽電子放射断層撮像法(PET: Positron Emission Tomography)などの手法を用いて明らかにしようとしている。また、メタ分析 (meta-analysis)、すなわち複数のイメージング研究の結果の俯瞰的な視点からの分析が行われている。Binder他は[21]、100以上の研究を集めたメタ分析を行ったところ、意味処理に関与する部位は68%が左半球で32%が右半球であった。また、一般的な意味処理(general semantic processing)に関与する部位として、大脳の左半球の3つの領域(後方の多種感覚の統合をおこなう連合皮質、多種感覚の統合をおこなう前頭前皮質内側辺縁領域)とそこに属する7つの部位(下頭頂葉後方、中側頭回紡錘状回海馬傍回下前頭回背内側前頭前野腹内側前頭前野後帯状回)が重要な役割を果たすとしている[21]

  • 後方の多種感覚の統合をおこなう連合皮質(posterior heteromodal association cortex)
1. 左下頭頂葉後方(posterior inferior parietal lobe; temporo-parietal junction, TPJ)は、多様な情報の統合と内的知識の検索をおこなっている[22][23][24][25]。具体的には、文や文脈の理解、問題解決、そして計画立案など、概念をうまく組み合わせるという役割を担っていると考えられている[21]
2. 左中側頭回(middle temporal gyrus, MTG)は、物やその属性に関する概念情報の蓄積をおこなっている[26][27][28][29][29][30][31][32][33][34][35][36][37][38][39][40][41]。一方、上側頭回はこれまで神経心理学の知見では言語理解の中心的な役割を担っていると考えられてきたが[13][42][43][44]、イメージング研究の知見ではことばの意味の検索というよりはむしろことばの知覚や音韻処理との関連が強いと考えられている[45][46][47][48][49][50][51]
3. 左紡錘状回(fusiform gyrus)は物の視覚的な属性に関する内的知識の検索に[52][29][53][54][55][56][57]左海馬傍回(parahippocampal gyrus)は外側の意味記憶と内側のエピソード記憶の仲介をしている可能性が示唆されている[58][59][60]
  • 多種感覚の統合をおこなう前頭前皮質(heteromodal prefrontal cortex)
4. 左下前頭回(inferior frontal gyrus, IFG)は、意味処理[61][62][63][64][65][66][67][53][68][69][70][71][72][73][74][75][64]、音韻処理や文法処理[76][77][78][64][79][80][67][81][82][83][72][23][73][38][48][84][85][86][87]に関与することが多数報告されている。左下前頭回内の詳細として、44野は音韻処理、45野と44野は文法処理、そして47野と45野は意味処理に関与することが示唆されている[88]
5. 左背内側前頭前野(dorsomedial prefrontal cortex, dmPFC)は、動き、注意動機づけの制御に関与している[89]。また、前方(anterior rostral medial frontal cortex, arMFC)はメンタライジング(mentalizing)の関与、後方(posterior rostral medial frontal cortex, prMFC)は不整合や間違いの監視に関与することが知られている[90]
6. 左腹内側前頭前野(ventromedial prefrontal cortex, vmPFC)は、動機づけ、感情、報酬の処理に関与しており、概念の情動的側面の処理の重要な役割を担っていると考えられている[91][92][93][94][95][90]
  • 内側辺縁領域(medial limbic regions)
7. 左後帯状回(posterior cingulate gyrus)は、エピソード記憶や空間視覚に関する記憶に関与しており[96][97][98][99][100][101][11][102][103][104][105][106]、未来の行動の参考とするために過去の経験の記録をしている可能性が示唆されている[21]

 大脳の右半球に関して、語用論の対象である比喩理解において右下前頭回の関与が報告されている[107][108]。一方、皮肉理解における右半球の関与に関しては報告により多様である[107][109][110][111][112]。また、比喩理解や皮肉理解においては、尾状核扁桃体といった皮質下領域の関与も報告されている[112]

 このように、イメージング研究の知見は、上記の1. 左下頭頂葉後方と4. 左下前頭回は、環シルビウス溝言語領域に位置しており、前者は情報統合や内的知識の検索に、後者は音韻・文法・意味処理に関与していることから、そこが言語中枢であることを支持している。また、上記の2. 左中側頭回、3. 左紡錘状回と左海馬傍回は、環・環シルビウス溝言語領域に位置しており、意味記憶とエピソード記憶に関与していることから、これらは意味論的な情報の充填に関与していることを支持している。さらに、5. 左背内側前頭前野、6. 左腹内側前頭前野、そして7. 左後帯状回は、順に注意や動機づけの制御、感情や報酬処理、そしてエピソード記憶の記録に関与していることから、語用論的な情報の充填に関与していることが示唆されている。なお、語用論的な情報の充填に関しては、右半球言語領域や皮質下領域の関与も示唆されているものの、一貫した見解は得られていない。
