英:thalamic gating  (狭義:sensory gating of thalamus)

 末梢からの感覚情報が視床を経由する際に、上位中枢である大脳皮質へ送る情報を選択する機構。抑制性細胞の集合体である視床網様核が中心的な役割を担っていると考えられる。感覚系視床において主に取り上げられることが多い概念だが、運動系視床も基本的に同じ構造の回路を持ち、同様のゲート機構と提案されている。

視床ゲート機構とは

 嗅覚を除く感覚系の情報は視床(例:外側膝状体視覚)、内側膝状体聴覚))を経由して大脳皮質一次感覚野へと送られる。この際、すべての情報がそのまま伝達されるわけではなく、視床において情報が選別されている。Crickが提唱した「サーチライト仮説」は視床ゲート機構に果たす視床網様核の役割を分かりやすく説明しており[1]図1)、視床網様核からの抑制性入力によって、大脳皮質へ送る情報と送らない情報を選別する機構があると考えられている。

 視床ゲート機構に関する回路を初期感覚系に絞って単純化すると図2のようになる。視床網様核には抑制性細胞のみが存在しており、大脳皮質へ投射する視床細胞へ抑制性入力(図2点線)を与えている。視床網様核の細胞は、視床中継細胞の軸索側枝と大脳皮質6層の細胞の軸索側枝から興奮性入力を受ける[2]図2)。

 感覚系、特に視覚系においては古くからこの回路に着目し、視床網様核を中心とした抑制系が大脳皮質への情報のゲート機構を果たしている可能性が示唆されてきた[3][4]。感覚系のみならず、運動系視床においても視床網様核を介した同様の機構が存在する[5][6]。さらに、マカクサルを用いた解剖学的研究において前頭前野からも視床網様核が投射を受けることが分かっており[7]、感覚・運動系のみならず、注意などに関わる前頭前野も視床網様核を介して視床ゲート機構を制御していると考えられる。    こうした視床ゲート機構を作り出すには、視床と同様、視床網様核もトポグラフィーを持ち、さらに視床網様核が視床に対して側方抑制(lateral inhibition)を与えていることが必要である。それらを支持する回路基盤がトポグラフィー[6][8][9][10][11]、側方抑制[10][11][12][13]図3A)の両面で示されてきた。また、視床と視床網様核の間には反回抑制の回路(図3B)もあり、これらは睡眠―覚醒時の視床の神経活動の同期性を調節することにより、大脳皮質への情報の伝達へ重要な役割を果たしている可能性がある[14]

具体例

視覚

 視床において視覚的「注意」が向いた対象の信号を増強し、それ以外の信号を抑制することが報告されている[15][16][17][18]。視床網様核の働きにより、「注意」が向いている視野に受容野を持つ視床外側膝状体の神経細胞の活動は上昇し、その周囲、すなわち「注意」が向いていない部位に受容野を持つ神経細胞の活動は減弱する[16]。このことによって「注意」を向けた対象物の視覚情報を選択的に大脳一次視覚野に伝達することができる。

体性感覚

 ヒト体性感覚視床である後腹側核(ventral posterior nucleus)領域に梗塞が起きた患者において、視床ゲート機構の障害が観察されたという報告がある[19]

聴覚

 げっ歯類ラット動物実験において、特異な音に注意を払うといった聴覚的「注意」によって視床網様核を介して内側膝状体の活動が制御されることが知られている[20]。ヒトでは統合失調症患者において、視床ゲート機構の機能低下あるいは消失が聴覚に関して報告されている[21][22]

痛覚

 内臓痛に関連して視床ゲート機構が働いているという報告がある[23]。また、麻酔時や睡眠時の感覚入力遮断にも視床ゲート機構が重要な働きを担っている[24][25]

複数の感覚間での視床ゲート機構

 視床網様核の神経細胞には、視覚、聴覚といった単一モダリティにのみ選択性をもつものだけでなく、例えば視覚に対して応答する細胞が聴覚に対しても応答するように、複数の感覚モダリティの入力に反応する細胞があることが知られており[26][27][28]、感覚を跨いだゲート機構も有していると考えられる。
