 錐体細胞とは、主に大脳皮質に存在する投射型興奮性神経細胞である。細胞体は錐形で、脳表面に向かう尖端樹状突起と細胞体近辺に伸びる基底樹状突起に棘突起が豊富に分布している。軸索の投射様式が異なるサブタイプから成り、大脳皮質の領野内・領野間及び、皮質から皮質下への情報伝達に重要な役割を果たしている。サブタイプ間で形態的・生理的・遺伝子的特徴やシナプス結合にも違いが見られ、これらは機能的に異なる役割を持つ。

定義

 錐体細胞は主に脊椎動物中枢神経系に存在し、哺乳類においては大脳皮質や海馬などに分布する興奮性の神経細胞である。細胞体が錐形をしていることに由来し、錐体路(pyramidal tract)とは名称の由来が異なる。細胞体は直径20-70 μm程であり、神経伝達物質としてグルタミン酸を使う。軸索は遠距離に投射する。スペインの神経解剖学者Santiago Ramón y Cajalらによる一連の研究により、詳細な形態が明らかにされた。

 対義語として、非錐体細胞と呼ばれる神経細胞があり、細胞体は楕円ないし円形で尖端樹状突起基底樹状突起の区別がない。ほとんどの非錐体細胞は皮質下には投射しない。非錐体細胞は、典型的にはGABA作動性の抑制性介在細胞を指すことが多いが、後述する有棘星状細胞 (spiny stellate cell)などの興奮性細胞もこう呼ばれることがある。

 網膜視細胞である錐体細胞(cone cell)についてはここでは記載しない。

解剖学的な特徴

5層に錐体の細胞体があり、1層に向けて尖端樹状突起を伸ばしている。尖端樹状突起の末端は房状突起となって終わる。細胞体の基部からは細い軸索と複数の基底樹状突起が伸びている。

 典型的な錐体細胞では、尖端樹状突起(apical dendrite)と呼ばれる一本の樹状突起が錐体形をした細胞体の頂点から生じ、斜め方向に分枝(oblique branch)を出しながら、数百μmに渡って一方向に直線的に伸びる (図1)。多くの場合、oblique branchはあまり分岐を繰り返さない。尖端樹状突起の末端は、房状分枝(tuft)を形成して終わることが多い。また、数本の基底樹状突起(basal dendrite)が、細胞体の底辺部から伸び、細胞体周辺の比較的限局した領域で分枝を出し、全体として半球ないし球状(半径およそ300μm)に広がる。樹状突起は棘突起を豊富に持ち(錐体細胞1個当たりの棘突起数は海馬CA1で約30000個、大脳皮質視覚野で約15000個と推定されている[1])、棘突起の形状や大きさ、安定性は学習の影響を受けることが知られている[2]。興奮性入力の多くは、他の錐体細胞や視床に由来し、シナプスは主に棘突起上に形成される。他方、抑制性入力は細胞体や樹状突起の幹に入力する割合が高い。以下、脳内の各部位における錐体細胞について述べる。

扁桃体

 扁桃体基底外側核群のおよそ90%が錐体細胞であり、大脳皮質や線条体などへ投射する。後述する大脳皮質の錐体細胞と似た形態であるが、尖端樹状突起と基底樹状突起の区別のはっきりしない有棘星状細胞(spiny stellate cell)様のものもある。尖端樹状突起の方向性は厳密ではないが、吻側方向へ向かう傾向がある[3]

海馬

 海馬[4]の錐体細胞はアンモン角(Cornu Ammonis, CA)の錐体細胞層(厚みは細胞体5個程度)に限局して存在する。アンモン角は錐体細胞の形態からCA1CA3の小領域に分けられる。CA3からCA1にかけて、細胞体の大きさは小さくなり、尖端樹状突起は細くなる傾向がある。細胞体は基部で幅20-40μm、高さが40-60μmの錐形であり、尖端樹状突起は放線層を経て網状分子層へと海馬の中心方向に向かって伸びる。CA3錐体細胞には棘状瘤(thorny excrescence)と呼ばれる巨大な棘突起が存在し、苔状繊維(mossy fiber)からの入力を受けている。CA3錐体細胞は、反回側枝(recurrent collateral)により互いに神経結合しており、CA2,CA1領域へはシャッファー側枝 (Schaffer側枝)と呼ばれる軸索を伸ばす。一方、CA1錐体細胞は海馬台(subiculum)や嗅内野(entorhinal cortex)へ投射している。

大脳新皮質

 6層構造の新皮質では、1層を除いて、各層ごとに特徴的な形態を持つ錐体細胞が存在する[5]。軸索はミエリン化しており、白質方向へ延びていきながら局所的な分枝を出し、これにより近隣の細胞群と局所回路を形成する。このような局所的な結合や皮質下への軸索投射は、層構造に関連があることが知られている(表)。

 1層は非錐体細胞で占められている。

 2層の錐体細胞は比較的小さく、尖端樹状突起は細胞体の近くで分岐しtuftを形成する。その軸索は主として皮質内結合や半球間結合に関わることが知られている。

 3層は中型の錐体細胞を含み、皮質間・半球間投射に主に関与する。

 感覚野の4層には、有棘星状細胞や尖端樹状突起の発達の乏しい細胞(Star pyramidal cellとも呼ばれる)が多く存在し、主として皮質内投射に寄与する。

 5層の錐体細胞は、大きな細胞体と太い尖端樹状突起を持ち、様々な皮質内・皮質下領域へ投射している。一次運動野5層には、特に巨大なBetz細胞(細胞体直径が100μmにも達する)が存在し、その軸策は錐体路(皮質脊髄投射)を形成する[6]

 6層の錐体細胞は、比較的小さな細胞体と細い尖端樹状突起を持ち、主として視床へ投射する。

 また、5層深部と6層にはmodified pyramidal cellと呼ばれる非典型的な形態の錐体細胞が比較的多く観察され、その尖端樹状突起が伸びる方向は細胞によって様々である。しかし、棘突起の存在や興奮性のシナプス結合など、多くの特徴を錐体細胞と共有している[7]

 1層には錐体投射細胞は存在しないので省略したが、錐体細胞の尖端樹状突起が密に分岐しており、シナプス結合そのものは豊富である。
層間結合・投射先共に、皮質領野によって違いがみられる。
* 結合の同定には様々な手法があり、相互に矛盾しない結果が必ずしも得られているわけではないので、詳細については引用文献などを参照されたい[8] [9] [10] [11] [12]。錐体細胞のタイプによって、入出力関係に差があることも報告されている[10] [11]。層内の相互結合は全層で見られるので、表では省略した。

皮質錐体細胞間の結合

階層性結合パターンによってFF (feedforward)・FB(feedback)・lateral connection に分けられ、それぞれに特徴的な層分布が見られる。三角形は出力細胞の分布を示し、投射繊維の密度は黒白の濃淡で示した。

 錐体細胞間の興奮性結合は、機能性カラム内や単一の領野内での局所的な結合(local/ intrinsic connection)と、機能性カラム間・領野間・大脳半球間などの長距離の結合(long range/ extrinsic/ inter-areal connection)とに分けることができる。

 局所的結合については、層間の結合様式[8] [9] [10] (表)に加え、細胞レベルでの詳細な結合パターンが明らかにされてきている。同じ投射先を持つ錐体細胞同士や、感覚入力に対する応答性を共有する錐体細胞同士では局所結合の確率が高い[11] [12] [13] [14]視覚野では、同じ刺激反応特性を示す互いに離れた(0.5-数mm)機能性カラム同士が、錐体細胞の軸索によって相互に結合している[15] [16] [17]

 皮質領野には機能的な階層性があり、感覚野を例に取ると、感覚刺激に対する応答が最初に現れる一次領野から高次領野に情報が運ばれていくに従い、刺激応答性が特異化すると考えられている[18] [19]。機能的階層性の観点から、領野間の結合様式は、低次から高次の階層へのfeedforward(FF)connection、高次から低次の階層へのfeedback(FB) connection、似たような階層間のlateral connectionに分けることができる[20]

 霊長類視覚野では、FF投射は主に2/3層の錐体細胞から起こり、標的領野の4層にクラスター状の終末を形成する[21]。これに対し、FB投射は4層以外の錐体細胞から起こり、標的領野では4層以外の層へ拡散した終末を形成する[21]。出力細胞・入力線維の層分布は領野間結合ごとに固有のパターンがあり、多様なタイプの錐体細胞が関わることが示唆されている[22] [23] [24] [25] [26] [27] [28]。FF投射・FB投射のどちらにおいても、複数の領野へ投射する錐体細胞は少なく[29] [30] [31]、加えて、一つの錐体細胞がFF投射とFB投射の両方に関わることはほとんどないことが報告されている[32]。また半球間結合や高次脳領野間結合では、結合線維はlateral connectionに特徴的な皮質全層にまたがるカラム様の終末を作る[20](図2)。

 上述のような階層的領野間結合は他の感覚領野や動物種でも見られるが、層選択性などについては差異がある[32] [33] [34] [35]。新しい細胞ラベル法やOptogeneticsの応用により、感覚野-運動野間のような機能的に異なる領野にまたがる投射も含め、長距離結合に関する知見はさらに深められつつある[36]

電気生理的特性 (大脳皮質錐体細胞について)

 大脳皮質の錐体細胞は、脳スライス標本(in vitro)やin vivo条件の電気生理記録から得られた発火パターンから、いくつかのサブタイプに分けられてきた[38] [39] [40] (図3)。

 最も多く見られるのは、脱分極パルスに対して等間隔で規則的に発火するregular spiking(RS) 細胞であり、Mountcastleによって名づけられた[41]。RS細胞は、閾値以上の電流注入に対して持続的な反復発火で応答し、その注入電流と発火頻度は線形に相関していることから[42] [43]、細胞への入力の時間情報などを運ぶのに適すると考えられる。持続通電中に発火頻度が徐々に落ちる適応(accommodation)の程度によってさらにサブグループに分けられることもある[40] [44] [45]

 Intrinsically Bursting(IB)細胞は、脱分極パルスに対して高頻度で連続発火し[38]、特に閾値より少し上の電流注入に対しては顕著な脱分極に乗った3-5発のバースト発火(約200Hz)を示すのが特徴である。

 Fast rhythmic bursting (FRB) 細胞は、通電に対して短いinter-burst intervalで2-5発(200-600Hz)のスパイクから為るバーストを発射しChattering neuronとも呼ばれる[46]。持続的な脱分極状態の時は、20-80Hzの律動的なバーストとなる[46] [47]。個々のスパイクは小さな後過分極(afterhyperpolarization, AHP)とそれに続く後脱分極(afterdepolarization, ADP)の要素を含む。

 このような発火パターンは、細胞膜上のNa, K, Ca等のイオンチャネルの発現を反映している。IB細胞やFRB細胞においては、注入電流と発火頻度の関係は非線形な相関であり入力-出力の関係はRS細胞とは異なる。

 電気生理的特性は、他の特徴とも関係していることが知られている。上述の三種の錐体細胞は大脳皮質2-6層に存在するが、IB細胞は5層に多く[48] [49]、FRB細胞は2-4層に多い。5層のIB細胞は同層のRS細胞と比べて細胞体が大きく、樹状突起を広く伸ばしており、軸索は5-6層で分枝する。これに対して、RS細胞は細胞体や樹状突起の分枝範囲が小さく、軸索は1-4層で分枝する[50]。皮質内・皮質下から皮質へ層特異的な入力があることから、各サブタイプの錐体細胞は異なる入力を受ける可能性がある。また、RS細胞とIB細胞は皮質内で構成する神経回路や投射先が異なる[51] [52] [53]。錐体細胞の投射先や形態、発火様式などの特徴はそれぞれに相関しており、各サブタイプの入出力特性の違いや結合特異性の差異から、これらのサブタイプは機能的に異なる役割を果たすと考えられている。

発生・分化

 大脳皮質のGABA作動性の非錐体細胞が、主に大脳基底核原基に起源をもつのに対し、錐体細胞は脳室帯由来である。脳室帯で分裂・分化した神経芽細胞放射状移動により、軟膜方向へ移動し、神経細胞へ分化する。この過程で、発生時期の早い細胞ほど脳の深い側に分布する(inside-out) [54]。また、扁桃体の基底外側核群の錐体細胞は、腹側外套の脳室帯由来と考えられる[55]。近年、大脳皮質の錐体細胞の投射先などの性質が、転写因子や細胞外シグナル分子などの空間・時間的な発現によって制御されていることが報告されており、錐体細胞のサブタイプの分化に関する研究が進んでいる[56]
